INSIGHTS INTO THE PRODUCTIONS OF KERATINASE FROM STREPTOMYCES BADIES VAR. SHASHI

Abstract

Abstract Keratins are fibrous and hard-degrading proteins, insoluble in water and organic solvents, often accumulated in nature and it is found majorly in hair, skins, feathers, hoofs, horn and nail etc.Keratinases are proteolytic enzymes having ability to degrade rigid and insoluble keratinous proteins.Keratinases are found in wide sources like archaea, bacteria, actinomycetes and fungi. Keratinase are mostly serine proteases but very few reports showing metallokeratinases. Several thermophile bacteria know to produce keratinase which can degrade keratin at ≥70°C.Keratinase activity found over wide range of conditions which would be useful in biorecycling of keratin wastes into feed and fertilizers.Keratinase has important applications in various industries such as leather, cosmetic, textile, biomedical and detergent industries, however recently it has been used in energy generation and green synthesis of nanoparticles. In the present study we have isolated 22 actinomycets from soil which showed ability to degrade keratin. Further biochemical tests were performed for initial identification. Further all the species were subjected to check its keratinolytic activity. Out of all the species the higherkeratinolytic activity was showed byStreptomyces badies, Srteptomycesparvus and Streptomyces graminerus. Further the species was identified by 16SrRNA sequencing which showed that Streptomyces badies var. Shashi is most prominent keratinase producing species.Keratinase enzyme was produced in minimal medium and then it was partial purified. Further the purified keratinase can be used in various industrial applications.
