Bacterial are well known for their ability to excrete enzymes into the environment. Microbial proteases play an imperative task in various biotechnological processes. Screening and isolation of protease producing strains of bacteria were carried out from soil collected near dairy in Virar. The isolates that were positive on skim milk agar (1%) were selected as protease producing strain. The organisms were tested for various biochemical tests, which lead to their tentative identification as Bacillus species producing protease enzyme. Enzyme production was carried in 100ml of production media in the fermenter at 37ºC for 48 hours at pH 8.0. Harvested protease product was partially purified by salt precipitation method and dialysed. Then protein was further characterized using SDS-PAGE.
