Title

DESIGN, FORMULATION AND EVALUATION OF BILAYER TABLETS GLIMIPIRIDE AS IMMEDIATE RELEASE AND ROSIGLITAZONE AS EXTENDED RELEASE

Abstract

The purpose of the present study was to design an bilayered tablet consisting of glimipiride core tablet for immediate release to produce immediate therapeutic effect, which is a layer on rosiglitazone for extended release of rosiglitazone. The tablets were designed using disintegrants for extended release and outer layer was designed as immediate release formulations using polymers Kollidon SR,HPMC K4M ,Ethocel to modulate the drug release.The invitro dissolution kinetics followed Kosmeyars-Peppas model via a non-fickian type diffusion controlled release mechanism for extended release. IR study reveals that there were no interaction between the drug and polymers used in designing of bi-layered tablets. All the rate dat (K) were found to be significant at the level of P<or=0.05 assessed.The in-vitro release profile shows the desired biphasic release behaviour. The glimepiride was dissolved within 30 mins, where as the rosiglitazone was able to release more than 12 hrs from formulations and t1/2were found to be 6.3hrs, and 6.9hrs from the formulations F2 and F4 respectively, which indicate the suitability for patient compliance.
