Title

Lipase in germinated Sunflower (Helianthus annuus) seeds: extraction, immobilization and partial characterization

Abstract

Lipase enzyme is used extensively as biocatalyst in various industrial processing and formulation. In view of such importance many researches have tried to develop reliable and simple method for optimization of lipase from different sources. In present study we partially purified and characterized lipase from germinating sunflower seeds. Lipase extracted from germinated seeds was partially purified by using ammonium sulphate fractionations. Immobilization and characterization was performed by biochemical parameters. In case of fractionations, 90% ammonium sulphate saturation was effective for lipase purification from crude extract as compare to 30 and 60% saturation of same salt. Lipase was purified with of 2.36 fold and increasing specific activity of 0.71. Sunflower lipase could be successfully immobilized in calcium alginate beads. The optimum temperature of lipase was between 40 and 500C and the optimum pH of lipase was 8 and 9. CuSO4 exhibited inhibitory activity against lipase. Germinated sunflower seed is a rich source of lipase could be utilized for industrial processing.
