SPROUTING BEANS IN WATER WITH HERBAL EXTRACTS TO PREVENT BIOFILM FORMATION BY PATHOGENIC & SPOILAGE BACTERIA

Abstract

Abstract:A challenge for the food industry is a pathogen-free food with an acceptable shelf life. Bio-control approaches using plants extracts prevent biofilm formation or disrupt them and enhance the safety of food supply by preventing spoilage and bacterial infections. Eight herbal extracts were selected & evaluated for the inhibition of E.coli, S.enteritidis, S.aureus & B.subtilis. Using agar cup diffusion technique, Amla was found to be most effective on S.aureus and S.enteritidis, ajwain extract showed maximum zone of inhibition for S. enteritidis & B. subtilis. Tulsi extracts significantly inhibited E. coli along with others. MIC & MBC for all three extracts aamla, ajwain and tulsi; was found to be 5mg/ml or lower against all test organisms [except for amla extract against B. subtilis]. A combination of these three extracts was expected to show synergistic action. With even 1mg/ml mixture of each extract 10000 fold reduction in E. coli count was observed when exposed moong seeds were crushed and plated. The least reduction was of B. subtilis which was still 100 fold. Significant inhibition of biofilm formation was also observed. A combination of these three extracts in concentrations not affecting palatability and with some compromise on % germination can be used to effectively reduce or prevent biofilm formation by these spoilage and pathogenic bacteria on & in moong bean sprouts.
