Title

Isolation, Screening and Characterization of Amylase Producing Microorganisms from Different Soil Sources

Abstract

Amylases are amongst most widely used enzymes in industries such as food, fermentation, textile and paper. The aim of the present study was to isolate, screen, and characterize amylase producers from different soil sources and to carry out production through fermentation process. The crude enzyme was extracted to check its enzyme activity and the production medium was optimized for various parameters. The isolates exhibiting the maximum zone of clearance on starch agar media by iodine were identified. All the isolates were characterized morphologically and biochemically. Total 10 bacterial isolates and 5 fungal isolates showed amylolytic activity Fermentation of bacterial and fungal isolates was carried out for 48 and 72 hours respectively. Amylase activity was assayed using 3, 5-Dinitrosalicyclic acid (DNSA) method. Bacterial isolates B7 and fungal isolate F4 showed highest activity. The maximum amylase activity obtained by bacteria and fungi were 3.09 U/ml and 2.88 U/ml respectively. By optimizing various parameters for maximum amylase enzyme production, the present indicates that the amylase activity for bacterial and fungal isolates were obtained best at pH 7, temperature 37℃ and incubation time of 48 hours and at pH 7, temperature 28℃ and incubation time of 72 hours respectively.
