Title

CHARACTERIZATION OF AGARASE PRODUCING BACTERIUM ISOLATED FROM MARINE ECOSYSTEM OF SOUTH GUJARAT

Abstract

Marine ecosystem is the least explored ecosystem of the earth which contains microbes capable of producing different enzymes. This study was focused on characterization & purification of agarase producing organisms. In present study previously isolated organisms were screened on MMS agar medium. The isolate C-19 giving maximum zone was selected for further characterization study. The optimum pH & temperature for this enzyme obtained in study were 10 and 50ᵒC respectively. The effect of medium composition on agarase production was investigated in shake flask using ASW medium. The most suitable carbon source, nitrogen source, & salt source found were agar, yeast extract, calcium chloride respectively, the time course for the enzymatic reaction was obtained best at 35ᵒC at 60min.The partial purification of crude enzyme was performed by ammonium sulphate and good activity was found at 60% saturation. According to the Michaelis-Menton graph the Km for agarase was found 2400µg/ml and Vmax was 266.6µg/ml & according to Lineweaver-Burk plot Km was 2500µg/ml and Vmax was 285.7µg/ml. Enzyme activity after U.V mutation was obtained best at 40sec.
