Title

Isolation and Secondary screening of Antibiotic producing Actinomycetes from different soil samples

Abstract

Antibiotics are secondary metabolites produced by microorganisms. Actinomycetes are the top producers of antibiotics. In the current study, total seven samples were collected from agricultural soil and rhizosphere soil of medicinal plants Tulsi, Aloe-vera, Neem, and Ardusi of local region. All the samples were isolated by using Glycerol Casein agar (GCA) medium. Among seven soil samples, total twelve Actinomycetes were isolated and further characterized on the basis of their colonial characteristics and Gram’s staining. The isolates obtained were Gram positive, rod shaped filamentous bacteria. Out of 12 isolated Actinomycetes, 10 (83.33%) showed antimicrobial activities against the tested microorganisms and from which 4 isolates were selected for their wide spectrum of antibiotics. In this study, isolates A6, A7, A8 and A9 were found to inhibit all of the test organisms. In order to extract the active compound from isolates, fermentation was carried out on 4 isolates and the former was extracted by using acetone. Antimicrobial compound obtained from isolates was separated by using separatory funnel. The antimicrobial compounds obtained from A6 demonstrate broad spectrum antimicrobial activity against Bacillus cereus, Sallmonella typhi, Escherichia coli and least was shown by crude extract of isolate A7 against Sallmonella typhi.
