Title

STUDY OF PH AND TEMPERATURE ON THE ACTIVITY OF BETA GALACTOSIDASE ENZYME FROM FUNGI

Abstract

Beta galactosidase is one of the most important commercial enzyme. It is found in many organism right from bacteria, fungi, bugs, plants, and animals. Fungi have various industrial usage as source of enzyme, protein, antibiotics and even bioremediation clearing pollutants. This study was done to determine the stability of the beta galactosidase enzyme from isolated fungal strains at varied pH of 4, 5, 6, 7 and 8. Also, thermal stability was determined to find out the activity of the enzyme at varied temperature range 28°C, 37 °C, 45 °C, 50 °C and 55°C. The enzymes from all the three isolates were found to obey the same pattern of being most stable around pH 6 to 7. The activity decreased after pH 7. The enzymes from three fungal isolates were found to be stable between 37°C to 45°C. The higher temperature of 55°C saw most decreased activity among all the isolates. The results clearly show that even though the three strains produced beta galactosidase enzyme that were stable for long range of pH and temperature but activity decreased drastically at extreme pH as acidic or basic and lower temperature to very high temperature. The isolate PT2 has higher enzyme activity among all the isolates at all temperature range with step by decline with increasing temperature. However, enzyme from PT3 showed sharp decline in activity after 45°C. For pH all the isolates followed the same pattern.
