Title

“Isolation of different types of microbial community from chronic ear infection samples also with antimicrobial characterization”

Abstract

To investigate whether secondary metabolite levels in plant of Vindhaya region are comparable to reported from other region and how these plants extract can be used to treat challenge. To test we prepared extracts from all parts of plant. These plant extracts were rich in steroids, terpenoids, alkanoids. Available literatures were systematically reviewed and we came to conclusion that the plant at these specific regions has mostly comparable properties to that reported from other regions. In addition our finding suggest that rather than taking one type of extract or biochemical therapeutics from one plant, we need to have a combinatorial approach for best medicinal property. Isolation of microbes for the further research work we have prepared several culture media such as Sabouraud Dextrose Agar (SDA), Nutrient Agar, Mac-Conkey’s Agar, Blood Agar, Chocolate Agar, Yeast Nitrogen Based Medium (YNB) . For the estimation of antibiotic sensitivity test we have used Agar Diffusion Method in which the molten agar will mixed with a different concentrations of the test samples at molten state of 45-50°C and mixed aseptically with different amounts of Onion and Garlic extracts to a concentration of 10 µl, 20 µl, 40 µl, 60 µl and 80 µl which is equivalent to 5 mg/ml, 10 mg/ml, 15 mg/ml and 25 mg/ml of media. Then for the comparative and preliminary analysis of presence or absence of important secondary metabolites is carried out for extract of mixed plant part in three different solvents. All three tested solvents extracts in water, ethanol and methanol of Allium Sativum were rich in tannins, carbohydrates, amino acids and proteins. It has been found that Allium Sativum was rich in most of secondary metabolites except some special glycosides
