Title

formulation and evaluation of immediate release tablet of ilaprazole by using natural disintegrant

Abstract

Now a day’s advances in Novel Drug Delivery Systems (NDDS) with a goal to enhance safety and efficacy of drug molecule by formulating a suitable dosage form for administration and to attain improved patient compliance. The present work, immediate release tablets of Ilaprazole were planned with a view to enhance patient compliance by direct compression and sublimation method. The tablets were prepared by direct compression method, by using mucilage’s of hibiscus leaves, okra pod powder, guar gum as natural disintegrants along with directly compressible mannitol to enhance mouth feel. The immediate release tablets were also prepared by sublimation technique using natural and synthetic polymer such as gaur gum, Hibiscus leaves, Okra pod, and crospovidone. The prepared tablets were evaluated for hardness, friability, drug content uniformity, wetting time, water-absorption ratio and in-vitro dispersion time. Based on in-vitro dispersion time (approximately 25.23 seconds), three promising formulations (one from each disintegrants) were tested for in-vitro drug release pattern (in pH 6.8 phosphate buffers), stability studies (at 40º/75% RH for three months) and drug-excipient and interaction study were carried out (IR spectroscopy). Among the promising formulations, the formulation FH3 prepared by direct compression method (containing 9% w/w of hibiscus leaves and 20% w/w of microcrystalline cellulose) and in sublimation method formulation SCP9 (containing 9% w/w crospovidone and 4% w/w camphor as subliming agent) were emerged as overall best formulation based on drug release characteristics and in-vitro dispersion time of approximately 24.6s & 20.03s and t50% values of 3.3 & 2.55min compared to control formulation which has shown in-vitro dispersion time of 120s and t50% values of (>30min). Stability studies indicated that there are no significant changes in drug content and in-vitro dispersion time. IR spectroscopic studies indicated that there no drug interactions.
