Title

OPTIMIZING DRUG DELIVERY OF LOVASTATIN THROUGH EMULSION SOLVENT DIFFUSION USING NANOSPONGES

Abstract

In recent years, nanotechnology has emerged as a promising approach to drug delivery, offering improved drug efficacy, bioavailability, reduced toxicity, and better patient compliance. With the aim of formulating a novel drug delivery system, we developed nanosponges loaded with lovastatin using a variety of polymers such as Ethyl cellulose, Polyvinyl alcohol, βcyclodextrin, Pluronic F68, and Hydroxy Propyl β- cyclodextrin. To ensure the safety and compatibility of the drug-polymer combination, we conducted FTIR tests and found no interactions between the two. We then evaluated the resulting nanosponges for particle size, PDI, zeta potential, SEM, entrapment efficiency, and in vitro drug release. Our results showed that the particle size ranged from 295.5 to 578.8 nm, PDI ranged from 0.189 to 0.465, and zeta potential from -17.3 to -35.96 mV, with entrapment efficiency ranging from 78.38 to 95.77%. In terms of drug release, all nanosponges exhibited varying degrees of cumulative percentage release, with the highest drug release (96.60%) observed in the F6 formulation. Our release kinetic studies suggested that the release was first-order diffusion-controlled, and the n value (0.6017) from the Korsmeyer-Peppas model indicated a non-Fickian type of release mechanism. Overall, our study highlights the potential of nanosponges as an effective drug delivery system with the ability to target cells and tissues, thus offering a novel approach to pharmaceutical research.
