Title

Synthesis of Naproxen Based a New Chiral Reagent and Separation of Racemic Pharmaceuticals by Liquid Chromatography

Abstract

In this report, the (S)-naproxen-based activated chiral derivatizing reagents (CDR) were synthesized by coupling pentafluorophenol, and UV, IR, HRMS, NMR, and CHNS was used to characterized them. For the chiral recognition investigation, a racemic -blocker (atenolol) was selected for this study. The diastereomers of the chosen -blocker were prepared using highly reactive CDR under microwave heating conditions. RP-HPLC was used to separate the prepared diastereomers. The effect of varying mobile phase concentrations and sample concentrations was optimised on prepared diastereomers. The optimised structures were used to predict the elution order of diastereomers synthesised. LOD (0.208 ng mL−1), LOQ (0.624 ng mL−1), calibration range (0.01-1.0 mg mL−1), correlation coefficient, and recovery were the validation parameters for our method (99.54 percent for intra-day assay and 99.89 percent for inter-day assay).
