Title

DETERMINATION OF BATCH DOSE UNIFORMITY BY X-RAY POWDER DIFFRACTION

Abstract

This report is intended to determine the ‘Batch dosage uniformity by X-Ray powder diffraction technique’. It contains the importance of X-Ray powder diffraction technique in determining the batch dosage uniformity. XRD is an ideal method for analysing the phase composition in mixtures composed of multiple components. It is capable of quantitative analysis, providing precise identification and measurement of concentration of each phase. This method works by using diffracted X-Ray to directly observe the repeating atomic structure present in samples, making it an ideal tool for identifying and characterising these materials. The intensity pattern of each component in a mixture is directly proportional to the amount of that component present. A batch of popular anti- diabetic tablet ‘GLYCOMET GP 1’ was selected, which contain 1mg of Glimepiride and 500mg of Metformin. It is easily available in market, consumed by many populations and it helps control blood sugar levels thus prevent serious complication of diabetes, mainly used to treat type 2 diabetes mellitus. XRPD was performed for the selected batch of Glycomet GP1 tablet. The X-Ray diffractogram was plotted by using ORIGIN LAB software. It is the data analysis and graphing software. It offers advanced analysis tools, and apps for peak fitting, surface fitting, statistics and signal processing. The plotted diffractogram is compared with the reference diffractogram obtained from ICDD, CCDC, IUCr etc. Intensities were measured and calculated the ratio. From this batch dosage uniformity of selected batch of Glycomet GP1 was determined. Determining the batch dosage uniformity helps to ensure the strength of a therapeutic product remains within specified acceptance limits. It also ensures that all tablets are within a tolerance of their average weight giving intra and inter batch uniformity data.
