Title

Smart and Stimuli-Responsive Nanotechnology Platforms for Controlled Drug Release and Disease-Specific Targeting

Abstract

Stimuli-responsive nanocarriers represent the new frontier in precision drug delivery, enabling drugs to be released only upon exposure to internal (pH, enzymes, redox state) or external (temperature, light, magnetism) triggers. This manuscript provides a comprehensive analysis of smart nanocarriers, mechanisms of stimuli-response, material design considerations, and therapeutic implications. A comparison of stimuli-sensitive polymeric nanoparticles, metallic nanosystems, liposomes, and hybrid platforms demonstrates significant improvement in site-specific release, up to 70% increased intracellular drug concentration and reduced systemic toxicity. The findings highlight the potential of multi-stimuli-responsive systems for treating cancer, neurological disorders, and chronic inflammatory diseases.
