Title

ETHOSOME AS A TOOL FOR DERMAL AND TRANSDERMAL DELIVERY: CURRENT PROSPECTS AND APPLICATION

Abstract

Vesicular systems are gaining traction as highly effective drug delivery mechanisms, showing promising outcomes. Among the array of vesicular carriers, Ethosomes stand out as particularly successful for delivering drugs susceptible to first-pass metabolism and those causing gastrointestinal side effects. Ethosomes represent a new generation of elastic liposomes, offering advantages over traditional liposomes such as enhanced entrapment efficiency and smaller vesicle size. Composed primarily of phospholipids, alcohol, cholesterol, and edge activators, Ethosomes notably contain higher concentrations of ethanol. They are primarily administered topically and transdermally, offering a non-invasive route for drug delivery through the skin layers and into systemic circulation. Ethosomes can be formulated into various preparations, including gels, lotions, ointments, and patches. They are categorized into three main systems: classical Ethosomes, binary Ethosomes, and Transethosomes. Among these, Transethosomes are considered superior due to their enhanced stability and elasticity. The preparation of Ethosomes involves various methods, and their quality is evaluated based on parameters such as vesicle size and shape, Zeta potential, entrapment efficiency, and skin permeation. This review focuses on the components, preparation methods, evaluations, and different types of Ethosomes, along with patents related to this innovative drug delivery system.
