Title

Cryoconservation efforts for endangered orchids: preserving Dendrobium longicornu lindl. and Dendrobium formosum roxb. ex lindl.

Abstract

Cryopreservation plays a crucial role in conserving plant species and genetic resources, especially for endangered species like orchids. Orchid seeds are vital for propagation and conservation efforts, yet their high desiccation sensitivity and limited longevity pose challenges for long-term storage. Only a small fraction of orchid species have been successfully cryopreserved, with a mere 0.34% success rate reported. In this study, Dendrobium longicornu Lindl. and Dendrobium formosum Roxb. ex Lindl., were used for cropreservation. Seeds proved to be the most suitable, with vitrification and encapsulation-dehydration methods applied. Vitrification resulted in 39.44% germination for D. longicornu and 40.77% for D. formosum after an hour in liquid nitrogen. The encapsulation-dehydration method yielded 24.7% germination for D. longicorum and 24.04% for D. formosum under specific sucrose treatments and dehydration conditions. PVS2 emerged as an effective cryoprotectant for both dendrobe species. The cryopreserved seeds, when cultured, developed into plantlets identical to their untreated counterparts, showcasing the potential of cryopreservation in preserving orchid biodiversity.
