Title

TRANSETHOSOMES: NOVEL VESICULAR CARRIER FOR ENHANCED TRANSDERMAL DRUG DELIVERY SYSTEM

Abstract

Transdermal drug delivery has emerged as a promising method for administering drugs systemically, offering benefits like non-invasiveness, controlled release, and bypassing first-pass metabolism. However, traditional transdermal systems encounter issues such as low bioavailability and restricted skin barrier permeability. Recently, transethosomes have garnered attention as an innovative drug delivery system aimed at enhancing transdermal drug delivery effectiveness. This paper presents a comprehensive overview of transethosomes, encompassing their formulation, key components, preparation techniques, characterization methods, advantages, applications, challenges, and prospects. The formulation of transethosomes involves blending phospholipids, ethanol, and edge activators or permeation enhancers to optimize drug delivery efficiency. Various preparation techniques, including thin-film hydration, and cold, hot, and ethanol injection methods, offer versatility in customizing transethosome formulations to meet specific therapeutic requirements. Characterization methods such as size analysis, morphology examination, and stability testing ensure the quality and functionality of transethosomes. Transethosomes offer numerous advantages, including improved permeation, sustained release, and enhanced flexibility, making them suitable for delivering a broad spectrum of drugs across the skin barrier. Clinical research has showcased their potential in diverse therapeutic areas, encompassing cancer therapy, infectious diseases, chronic ailments, and immunotherapy. However, challenges like scalability issues, manufacturing intricacies, and regulatory considerations need addressing to facilitate their clinical adoption. Transethosomes represent a significant leap forward in transdermal drug delivery, offering a promising solution to the limitations of conventional delivery systems. Through ongoing research and innovation, transethosomes have the potential to transform drug delivery and enhance patient outcomes across a myriad of medical conditions.
