Title

Method development and validation of a stable HPLC method to measure benazepril hydrochloride in both bulk and Pharmaceutical dosage forms

Abstract

Benazepril is an angiotensin-converting enzyme (ACE) inhibitor, which is commonly used in the treatment of diabetes mellitus. In this study, a rapid high-performance liquid chromatography (HPLC) method was developed to quantify benazil in pharmaceutical dosage forms, and its compliance with FDA and ICH requirements was validated. A 75:25 v/v ratio of acetonitrile to water with 1% acetic acid was used as the mobile phase to create the best chromatographic conditions for the entire study. The linearity research results demonstrated a linear association between concentrations in the 40-1g/ml range. The accuracy findings indicated that the percentage recovery at all three levels was between 90 and 110%, and the percentage RDS values were between 0.6 and 1.8%. The percentage recovery and percentage RSD readings, which were within the allowed parameters of 90% to 110% and not more than 2.0%, respectively, demonstrated the method's suitability for regular drug analysis. Using the systematic approach outlined in Section 2.6.6, by analyzing the impact of minor changes to the HPLC settings on the system suitability parameters, the robustness of the proposed technique was evaluated. Under both conditions, the sample and standard solutions showed good stability over a 24-hour period, as evidenced by the recovery percentage falling between 98.0% and 102.0%. It was also demonstrated that the tailing factor and the theoretical plate count were both within acceptable bounds.
