Title

DETERMINATION OF PRODUCT SHELF LIFE AND EVALUATION OF KINETIC PARAMETERS OF THERMAL DECOMPOSITION OF OXYCLOZANIDE IN SOLID STATE BY HPLC.

Abstract

A new high-performance liquid chromatography (HPLC) method for the quantitation of Oxyclozanide has been developed and validated. Oxyclozanide was subjected to thermal decomposition at 140, 150, 160, 170, 175,180 and 190°C. Samples were withdrawn at intervals of 7, 14, 21 and 28 days and analyzed for remaining drug content by HPLC assay method. The samples were eluted on a Cosmosil C18 column (250 mm x 4.6 mm, 5.0 μm) at 25°C, with a mobile phase of 3% of glacial acetic acid in Methanol : Water :: 97 : 3, (v/v). The flow rate was 1.0 mL/min, Injection volume: 20µL and detection by PDA detector with detection wavelength at 300 nm. Oxyclozanide shows thermal-degradation after exposure to high temperature. The shelf life of Oxyclozanide was determined by accelerated stability studies on the basis of first order degradation kinetics. The reaction rate constants, activation energy and half life were also calculated using Arrhenius equation and plots. The shelf life of Oxyclozanide was found to be approximately (t90 ) 6.36 years. Amounts of Oxyclozanide in all samples were determined by HPLC and the results were compared with those from Infra red spectroscopy and X-ray diffraction technology for qualitative comparison.
