Title

REVERSE PHASE HPLC METHOD DEVELOPMENT AND VALIDATION FOR SIMULTANEOUS ESTIMATION OF GLIMEPIRIDE AND LINAGLIPTIN

Abstract

A simple, rapid, precise, accurate, economical and reproducible reverse phase high performance liquid chromatographic (RP-HPLC) method was developed and validated for the determination of Glimepiride (GLM) and Linagliptin (LIN) that is used in combined ratio of (2: 5) for treatment of patient suffering with Diabetes.[1] The separation was achieved on a Shim-pack solar C18 (250 mm × 4.6 mm, particle size 5 µm) using a mobile phase [Acetonitrile: Methanol: Water (pH 3.0 adjusted with 1% (OPA) Orthophosphoric Acid)] (80:10:10 %v/v/v). HPLC separation of GLM and LIN was carried out at the detection wavelength of 228 nm. The mobile phase was pumped at a flow rate of 0.8 ml/min. The retention times of GLM and LIN were found to be 4.970 min and 2.451 min respectively. Excellent linearity range was found between 2-6 µg/ml for GLM and 5-15 µg/ml for LIN. The method was validated with respect to linearity, precision, accuracy, specificity and ruggedness. Method was successfully applied for the simultaneous determination of Glimepiride (GLM) and Linagliptin (LIN) in combined pharmaceutical dosage form synthetic mixture.
