Title

AN INSIGHT INTO ENGINEERING STIMULI-RESPONSIVE NANOPARTICLES FOR TARGETED DRUG DELIVERY IN CANCER THERAPY.

Abstract

For the past several years, stimuli-responsive nanoparticles (SRNPs) have attracted much attention as promising platforms for targeted drug delivery in cancer therapy. Based on a series of engineering approaches and targeting mechanisms regarding SRNPs, this article provides a detailed overview of in vitro and in vivo research. In the first part, it discusses the problems associated with conventional treatments of cancers and presents potential opportunities in overcoming these problems by SRNPs. This thorough review covers various types of SRNPs, design principles, and some progresses in Nano-medicine. The targeting mechanisms include passively targeting and actively targeting mechanisms, and engineering tactics regarding material selection and surface functionalization are discussed. Some studies are critically reviewed concerning the safety and therapeutic efficacy of SRNPs in vivo and in vitro. An analysis of what is left in the way of obstacles and potential directions for SRNP development rounds off the review. It will thus be handy for academics and physicians interested in using SRNPs for targeted drug delivery in the treatment of cancer as a general resource.
