Title

DEVELOPMENT AND VALIDATION OF UV SPECTROPHOTOMETRIC METHOD FOR SIMULTANEOUS ESTIMATION OF DAPAGLIFLOZIN AND TENELIGLIPTIN IN BULK DRUG AND PHARMACEUTICAL FORMULATION

Abstract

Two simple, accurate and precise simultaneous estimation methods were developed and validated to assess Dapagliflozin (DAP) and Teneligliptin (TEN) in bulk drug and pharmaceutical formulation. Method A: The zero-order derivative spectroscopic approach that has been devised comprises measuring the amplitude of the ratio spectra at 223 nm for DAP and 243 nm for TEN. The developed method used distilled water as a solvent. Beer's law complied in the concentration range of 4-20 μg/ml and 8-40 μg/ml for DAP and TEN respectively. Method B: The Q-absorbance ratio method involves measuring absorbance at 243 nm for TEN and 231 nm for DAP as iso-absorptive point of both drugs. The proposed approach revealed that the percentage assay fell between 99 to 100.0 % for DAP and 99.62 to 100.0 % for TEN in method A and 99.16 to 100 % for DAP and 99.58 to 100.0 % for TEN in method B. The analysis results have been statistically evaluated in compliance with ICH recommendations. Recovery studies have verified the developed procedure's accuracy as well as reproducibility. DAP and TEN were discovered to have LOD numbers of 0.17593 μg/ml and 0.47668 μg/ml for method A and 0.6047 μg/ml and 0.4766 μg/ml for method B respectively. DAP and TEN were found to have LOQ numbers of 0.53313 μg/ml and 1.44450 μg/ml for method A and 1.8141 μg/ml and 1.4445 for method B respectively.
