Arginase inhibitors have emerged as promising agents in cancer therapy, targeting the dysregulated arginine metabolism that supports tumor growth and immune evasion. By modulating the tumor microenvironment, these inhibitors restore immune function and enhance antitumor responses. Recent advancements in medicinal chemistry have led to the development of potent and selective arginase inhibitors, including small molecules, natural product derivatives, and peptidomimetics. Despite their potential, clinical translation is hindered by challenges such as off-target effects, toxicity, and suboptimal pharmacokinetics. This review provides a comprehensive analysis of the progress in arginase inhibitor development from 2019 to 2024, highlighting innovative strategies in drug design and addressing key hurdles. Emerging trends, including combination therapies and advanced delivery systems, are explored, offering insights into future directions for optimizing their therapeutic potential in oncology.
