Title

BIOPYTHON AND NGS ANALYSIS OF HUMAN MUTANT IN COMPLEX WITH NADPH AND AG881 (VORASIDENIB) INHIBITOR

Abstract

Abstract- This research explores the integration of computational tools and next-generation sequencing (NGS) data in analyzing human protein mutants, with a specific focus on the protein structure bound with NADPH and AG881 (vorasidenib) inhibitor. Using the protein data bank (PDB) structure with the code 6VEI, the study employs Genewise, InterProScan, Biopython, and STRING to uncover the molecular interactions, structural features, and biological pathways associated with this complex. Clinical research is being done on inhibitors of mutant isocitrate dehydrogenase (mIDH) 1 and 2 cancer-associated enzymes to treat a number of tumors with an IDH mutation. These inhibitors stop the buildup of the oncometabolite d-2-hydroxyglutarate (2-HG). Here, discovery of vorasidenib (AG-881), a strong oral dual inhibitor of mIDH1 and mIDH2 that penetrates the brain. Combined with computational tools like Biopython and Ngs facilitate efficient processing,analysis of data. By characterizing the drug binding site using X-ray cocrystal structures, we were able to comprehend the dual mutant inhibition.The results contribute to a better understanding of the mutant's role in disease pathogenesis and its potential as a therapeutic target.
