Title

INTRANASAL DRUG DELIVERY: ADVANCES, CHALLENGES, AND FUTURE PROSPECTS

Abstract

Intranasal drug delivery has emerged as a promising non-invasive route for administering a diverse array of therapeutic agents, particularly for treatments requiring rapid onset and direct central nervous system (CNS) targeting. This review delves into the latest advancements, challenges, and future directions of intranasal drug delivery systems. The nasal route offers distinct advantages, making it ideal for delivering both small molecules and complex biologics. The discussion includes various nasal formulations—liquid, semi-solid, and solid dosage forms—and emphasizes the impact of innovative drug delivery technologies like liposomes, microspheres, and nanoparticles in improving drug absorption and stability.Despite these advancements, several challenges persist due to the nasal mucosa's physical and enzymatic barriers, which can hinder drug bioavailability. Issues such as mucociliary clearance, enzymatic degradation, and environmental factors significantly affect the effectiveness of nasal drug delivery, necessitating meticulous formulation and design strategies. Additionally, the review highlights the regulatory and manufacturing complexities inherent in developing nasal drug products, underscoring the need for standardized protocols and collaborative efforts to address these challenges. Looking ahead, the integration of nanotechnology, smart delivery systems, and personalized medicine into intranasal formulations holds significant potential to expand the therapeutic scope of this administration route. As research progresses, intranasal drug delivery is poised to become a crucial component in the future of targeted, patient-friendly treatment strategies.
