The past five years (2020-2025) have witnessed transformative progress in cancer genetics, driven by an evolving understanding of the cancer genome and powerful technological innovations. Research has deepened insights into somatic and germline mutations, the dynamic interplay of epigenetic modifications, and the expanding functional roles of non-coding RNAs, including newly discovered ncRNA-encoded peptides. Breakthrough technologies such as single-cell sequencing, liquid biopsies, CRISPR-Cas gene editing, and artificial intelligence are revolutionizing discovery and providing unprecedented resolution. These advancements are increasingly translated into clinical practice, fueling precision oncology, enhancing immunogenomic approaches like CAR-T cell therapy and neoantigen vaccines, and offering new strategies to target the complex tumor microenvironment. While challenges in data integration, tumor heterogeneity, and therapeutic resistance persist, the field is poised for continued innovation, focusing on multi-omics integration and predictive, personalized medicine to improve patient outcomes.
