Title

Invitro pH Studies In Mentha piperita L. And Ocimum sanctum L. Using MS Medium

Abstract

Invitro pH studies refer to experiments conducted outside of a living organism, typically in a laboratory setting, to measure and analyze the acidity or alkalinity (pH) of substances under controlled conditions. This study explores the impact of varying pH conditions on the In vitro development of Mentha piperita L. and Ocimum sanctum L. using Murashige and Skoog (MS) medium. Explants from both species were cultured in media adjusted to pH values of 3.7, 4.7, 5.7, 6.7, and 7.7. Observations over a 25 day period revealed differential responses in callus formation and shoot multiplication. The highest survival and growth rates were observed at Ph 4.7 for both species, with Mentha piperita showing 80% survival and Ocimum sanctum 70%. In contrast, Ph 3.7 resulted in complete culture loss. These findings emphasize the critical role of medium pH in plant tissue culture, suggesting that mildly acidic conditions (pH 4.7) are optimal for in vitro propagation of these species.
