Title

Phytochemical Profiling, Antioxidant Activity, and Antimicrobial Efficacy of Vacuum-Dried Balanites aegyptiaca Juice Powder Against Clinically Relevant Bacterial Strains

Abstract

The present study investigates the phytochemical composition, antioxidant potential, and antimicrobial efficacy of vacuum-dried Balanites aegyptiaca juice powder, a traditionally used medicinal plant with underexplored scientific validation. Phytochemical screening revealed the presence of alkaloids, flavonoids, tannins, saponins, and phenolics, while physicochemical analysis confirmed acceptable levels of total ash (3.65% w/w), moisture content (6.3% w/w), and acidic pH (3.61), indicating formulation suitability. Heavy metal analysis showed concentrations of lead, cadmium, and arsenic well below USP safety thresholds, confirming the product's non-toxic nature. High-performance thin-layer chromatography (HPTLC) quantified gallic acid content at approximately 25% w/w, underscoring the antioxidant potential. The total phenolic content was found to be 390.8 mg GAE/g, while the hydrogen peroxide scavenging assay demonstrated an IC₅₀ of 42 µg/mL, comparable to standard ascorbic acid (38 µg/mL). Antimicrobial studies using agar well diffusion and MIC/MBC assays showed notable activity against Bacillus subtilis (32.5 mm inhibition zone) and Staphylococcus aureus, with MICs ranging from 6.25–12.5 mg/mL and MBCs between 25–100 mg/mL. These findings suggest that Balanites aegyptiaca juice powder, rich in polyphenolic content and free from toxic contaminants, possesses significant bioactive properties and holds promise as a natural therapeutic agent against oxidative stress and multidrug-resistant bacterial infections.
