Title

Lecithin Organogel: A Promising Topical Delivery System for Dithranol in the Management of Psoriasis

Abstract

Psoriasis is a chronic, immune-mediated skin disorder that demands the development of stable and patient-friendly topical formulations with sustained therapeutic action. Dithranol, though an effective antipsoriatic agent, is limited by poor solubility, instability, and skin irritation in conventional formulations. In the present study, dithranol-loaded lecithin organogels (Dith-LOs) were developed using isopropyl myristate as the oil phase, lecithin as the gelling agent, and poloxamer 407 as the stabilizer to enhance drug stability and skin permeation. Nine formulations (DT1–DT9) with varying lecithin concentrations were prepared and characterized for viscosity, pH, drug content, spreadability, zeta potential, particle size, stability, and in vitro release. The optimized formulation (DT5) demonstrated an ideal balance between viscosity (3167 ± 30.14 poise) and spreadability, with a uniform drug content of 96.47 ± 0.81% and pH 6.33 ± 0.28, ensuring skin compatibility. The mean particle size (190.8 nm) and zeta potential (−40.1 mV) of the optimized formulation indicated excellent colloidal stability. Transmission electron microscopy confirmed spherical and uniform vesicles. The in vitro release study revealed a sustained drug release of 27.23 ± 1.06% after 24 hours, compared to faster release from the pure drug suspension. These findings suggest that lecithin organogels provide a stable, biocompatible, and controlled-release vehicle for dithranol delivery, making them a promising platform for psoriasis management.
