Title

Evaluation of Antioxidant and Anti-Angiogenic Potential of Root and Stem Bark Extracts of Calotropis procera selected from Sariska Tiger reserve Alwar

Abstract

Oxidative stress and pathological angiogenesis play critical roles in the progression of chronic inflammatory disorders and cancer. The present study evaluated the antioxidant and anti-angiogenic potential of hydroalcoholic (70% v/v ethanol) extracts prepared from the root and stem bark of *Calotropis procera* collected from the Sariska Tiger Reserve, Alwar, Rajasthan. Extracts were obtained by cold maceration, yielding 12.40% for root and 16.80% for bark. Antioxidant activity was assessed using the 2,6-dichlorophenolindophenol (DCPIP) decolorization assay with ascorbic acid as standard. Both extracts exhibited concentration-dependent activity; the root extract showed superior potency (IC₅₀ = 185.6 µg/mL) compared to the bark extract (IC₅₀ = 238.4 µg/mL), while ascorbic acid displayed an IC₅₀ of 14.2 µg/mL. Anti-angiogenic activity was evaluated using the HUVEC tube formation assay on Matrigel. Root and bark extracts inhibited VEGF-induced tube formation in a dose-dependent manner, with IC₅₀ values of 132.4 ± 8.7 µg/mL and 212.6 ± 11.3 µg/mL, respectively. Suramin (standard) showed an IC₅₀ of 28.6 ± 2.1 µg/mL. Microscopic observations confirmed progressive disruption of capillary-like networks with increasing extract concentrations. The findings demonstrate that hydroalcoholic root extract of *C. procera* possesses stronger dual antioxidant and anti-angiogenic activity than the bark extract, supporting its potential as a multi-target phytotherapeutic agent against oxidative stress-related and angiogenesis-driven disorders.
