This study is an attempt to standardize an in vitro culture technique for propagation and conservation of Semecarpus anacardium L. Murashige and Skoog medium supplemented with 6-benzylaminopurine (BAP) and Kinetin (KIN) were used as culture media for proliferation of shoot from explant. For root induction, half strength MS medium supplemented with Naphthalene acetic acid (NAA) and Indole Butyric Acid (IBA). Maximum no. of shoots obtained was 57.8±5.9 per explant on MS medium supplemented with BAP at concentration 3.5mg/L with an average 96.9±2.8% rate of proliferation and the average length of shoot was 5.8±0.2 cm. The root generation rate is best (95±5.0%) at 0.75 mg/L concentration of NAA. No. of roots (9.7±1.9) and length of root (6.2±1.1cm) is maximum at this concentration of NAA. The regenerates of Semecarpus anacardium L. were adapted well in green house and in outdoor conditions afterwards.
