Title

Metagenomics and Next generation sequencing: a combinatorial approach

Abstract

India has a significant proven coal reserve appropriate for the CBM development. The bituminous and subbituminous rank of coals is being produced from the Jharia basin of Jharkhand being the largest producer of CBM in India. Deep coal deposits, which are not accessible by conventional mining operations, hence suggest suitable CBM development openings. Biogenic methanogenesis has been reported from the Jharia coal bed basin; although no sequential involvement of the microbes and the transformation of the coal organics to methane have been obtained. More accurate information regarding the microbial communities of biogenic CBM production is required. The use metagenomic approach via the 454 pyrosequencing, a widely referred sequencing technique with a high read accuracy rate has overcome the cloning bias and restricted throughput methodological limitations and can reveal more accurate microbial ecology data. The direct sequencing of the genomic DNA isolated from the samples shall help in bypassing the cloning bias and throughput restrictions.
